I'm assuming they need to create an account on the site? If so, require them to supply an email adress, and send an email to it with an activation code for their account. Only once they've actived the account, they can use it. If the email address supplied is invalid, they wont get the email, and thus they know they did something wrong, and you wont have invalid email adresses in your database :-) Of course, you'd have to delete the un-activated accounts if they're over a week old, cause that'd mean the user hasn't gotten their email, or they didn't bother to activate their account.
