Could be your version of mysql doesn't support MD5? Works for me - note the
version:
~$ mysql test
Welcome to the MySQL monitor. Commands end with ; or \g.
Your MySQL connection id is 15465 to server version: 3.23.12c-alpha
Type 'help' for help.
mysql> update test set description = md5(description);
Query OK, 4 rows affected (0.00 sec)

mysql> select * from test;
