When you install IIS it creates a user on your system called IUSR_<MACHINENAME> where <MACHINENAME> is the name of your computer - so on my desktop machine, the user is called IUSR_PHILSNTSERVER. This user (as you might expect!) has limited access to machine resources. If you want to change the user to something else, you can do that in the microsoft management console under the directory security tab in the properties dialog box for your website. If you want to change to a different user, make sure that the new user has 'log on locally' permissions.
