PHP can handle this (although you will definitely have to disable script timeout) in one run. But a smarter thing to do would be to not send all the 5000 emails at one time rather come up with a method that runs the PHP script every 5 minutes (a cron job) and send the newsletter out in 200+ email batches. This will reduce the load one the web server and also the mail server.
