I think that FAT is brain dead in respect of permissions, so it might be that IUSR_<machinename> doesn't have sufficient privileges to run php. Try temporarily changing the anonymous IIS user to an administrator and see if that cures things (in internet service manager go to anonymous access and authentication control in the directory security tab for your website's properties). If that fixes it, you need to either sort out privileges of IUSR_ or create a new user for the job.
