ZEND_DEBUG is a compile-time definition, that sets whether PHP is built with additional information, that may be helpful when debugging it. It's not useful for debugging PHP applications. Either way, you can't change it in runtime in any way, because setting it differently at compile-time results in different executables.
You can set it in compile-time using --enable-debug or --disable-debug (the latter being the default).
